Mucolipin 2 (MCOLN2)
TRPML2 is associated with the Arf6-regulated trafficking pathway and is involved in the intracellular transport of membranes and membrane proteins.The deduced 538-amino acid protein shares a high degree of similarity with MCOLN1 and MCOLN3 in the pore region, with conservation of an asp-asp (DD) motif predicted to determine ion specificity. Immunofluorescence analysis of transfected HeLa cells showed colocalization of fluorescence-tagged MCOLN2 with CD63, a marker for late endosomes/lysosomes. Lighter MCOLN2 staining was also observed at the plasma membrane and in tubular endosomes, which are involved in ARF6-regulated protein recycling via the clathrin-independent pathway. MCOLN2 colocalized with major histocompatability complex I (MHCI) and CD59 in tubular endosomes.
