Mouse Myrip targeted the type II PKA holoenzyme to a specific perinuclear region in insulin-secreting cells. Myrip also interacted with the Sec6 and Sec8 components of the exocyst complex, which controls protein trafficking and exocytosis.
The deduced 856-amino acid protein contains an N-terminal SLP homology domain (SHD) with 2 zinc finger motifs and a C-terminal myosin VA (MYO5A)-binding site. By database analysis, they identified the human ortholog, which encodes a deduced 859-amino acid protein that shares 79.7% identity with mouse Slac2c. RT-PCR analysis of mouse tissues revealed highest expression of Slac2c in brain, lung, and testis, with weaker expression in heart, skeletal muscle, and kidney, and no expression in spleen and liver.
