GRIM19 showed a punctate distribution and colocalized with endogenous mitochondrial proteins. Mutation analysis indicated that the N terminus of GRIM19 harbors the mitochondrial localization sequence.
The deduced 144-amino acid protein has a calculated molecular mass of 16 kD. Northern blot analysis detected GRIM19 in all adult and fetal tissues examined, with highest levels in adult heart, skeletal muscle, liver, kidney, and placenta, and in fetal liver, spleen, and bone marrow. SDS-PAGE of in vitro translated cDNA indicated that GRIM19 has an apparent molecular mass of about 16 kD. Immunofluorescence microscopy localized GRIM19 to the nucleoplasm of transfected HeLa cells, with some punctate cytoplasmic staining.
