Small guanosine triphosphatases, typified by the mammalian Ras proteins, play major roles in the regulation of numerous cellular pathways.These proteins, termed kappa-B-Ras1 and kappa-B-Ras2, interacted with the PEST domains of I-kappa-B-alpha and I-kappa-B-beta and decreased their rate of degradation. Human kappa-B-Ras1 is widely expressed and encodes a mRNA of 1.6 kb.the region of kappa-B-Ras that interacted with I-kappa-B-alpha and I-kappa-B-beta is localized to amino acids 58 to 93 and includes a switch II region of the potential GTP binding domain (amino acids 64 to 79 in kappa-B-Ras1). Kappa-B-Ras proteins inhibited NF-kappa-B activation by NIK and IKK-beta, suggesting that kappa-B-Ras-mediated inhibition of NF-kappa-B takes place either at the I-kappa-B phosphorylation step or further downstream.
