PYPAF5 associated with ASC, whereas PYPAF2 (NALP2), PYPAF3, PYPAF4 (NALP4), PYPAF6, and PYPAF5 lacking the PYD did not. Immunoblot and luciferase reporter analysis showed that PYPAF5 and ASC synergistically participated in activation of NFKB and procaspase-1. PYPAF5 belongs to a subgroup of NALP proteins that regulate NFKB and cytokine production.the predicted 892-amino acid NALP6 protein, which they called PYPAF5, has an N-terminal PYD, a central NACHT-type nucleotide-binding site domain, and a C-terminal domain containing at least 4 LRR motifs. Real-time PCR analysis of peripheral blood leukocytes detected expression of PYPAF5 predominantly in granulocytes, with some T-cell expression. Northern blot analysis revealed little expression in other tissues and cell lines examined.
