Guanine nucleotide exchange is essential for RAB GTPase activities in the regulation of intracellular trafficking, a process facilitated by guanine nucleotide exchange factors (GEFs). Iwasaki and Toyonaga (2000) determined that Aex3, a yeast GEF for the Rab3 GTPase and an ortholog of the human MADD protein, regulates neural activities via a second pathway through interactions with Cab1 (C terminus of Aex3-binding protein-1). Cab1 is a highly conserved, 425-amino acid protein with a neural protein counterpart, NPDC1, in mouse and human.The International Radiation Hybrid Mapping Consortium mapped the NPDC1 gene to chromosome 9
