Ozz assembled into an active E3 ligase complex, which the authors called Ozz-E3, in the presence of other ligase subunits. Beta-catenin was a target substrate of Ozz-E3 in vivo. The human protein has a similar size and structure. Northern blot analysis of mouse and human tissues showed expression of OZZ in skeletal muscle and heart, but not in pancreas, kidney, liver, lung, placenta, and brain. At day 14 to 15 of mouse embryonic development, when most muscles have formed but secondary myogenesis has not yet begun, Ozz accumulated near the sites of sarcomere assembly in growing myotubes. In primary myoblast cultures, Ozz localized predominantly at the tips of differentiating myofibers. Western blot analysis showed endogenous mouse Ozz at an apparent molecular mass of about 30 kD.
