Nucleotides are involved in numerous biochemical reactions and pathways within the cell as substrates, cofactors, and effectors. Nudix hydrolases, such as NUDT12, regulate the concentrations of individual nucleotides and of nucleotide ratios in response to changing circumstances.
NUDT12 contains a consensus Nudix motif and an ankyrin repeat in its N-terminal half and an NADH diphosphatase-like sequence in its C-terminal half. Following transfection into HeLa cells, NUDT12 was targeted to peroxisomes, and the C-terminal tripeptide PNL was required for peroxisome targeting. About 1% of the protein expressed by transfected insect cells was secreted into the medium
