PDRG1 is upregulated by UV radiation but downregulated by tumor suppressor p53 (TP53). PDRG1 was upregulated by UV radiation in a p53-positive breast cancer cell line (MCF-7) and in p53-negative lung (H1299) and prostate/bladder (TSUPr1) cancer cell lines. Studies of PDRG1 regulation in a colon cancer cell line (DLD1) with p53 inducible and repressed expression, combined with results from PDRG1 promoter studies, indicated that UV upregulation of PDRG1 occurs in a p53-independent manner and may be mediated via the PDRG1 promoter Oct1 sequence. Downregulation of PDRG1 promoter activity by p53 may be mediated via a putative head-to-tail response element. Cells from a human lung cancer cell line (HCT116) overexpressing PDRG1 displayed decreased survival following UV treatment.
