By sequencing clones obtained from a size-fractionated adult brain cDNA library, Nagase et al. (2000) cloned PHRF1. The deduced 1,654-amino acid protein shares significant similarity with the rat A9 protein, which contains a ser/arg-rich domain and binds the C-terminal domain of the RNA polymerase II large subunit (POLR2A). RT-PCR ELISA detected ubiquitous expression, with highest levels in ovary and adult and fetal liver. Strong expression was also detected in testis, brain, spleen, lung, skeletal muscle, heart, and pancreas, and moderate expression was detected in kidney. Expression was strong in all specific adult brain regions examined, with highest level in corpus callosum.By radiation hybrid analysis, Nagase et al. (2000) mapped the KIAA1542 gene to chromosome 11.
