PITPNM2 shares structural domains with other PITPNM family members, including an N-terminal phosphatidylinositol (PI) transfer domain, an acidic calcium-binding region, 6 transmembrane domains, and a conserved C-terminal domain.The calculated molecular mass of this variant is 125 kD. Northern blot analysis detected PITPNM2 transcripts of 7.5 and 9.5 kb that were highly expressed in thymus, heart, brain, ovary, and testis. Immunohistochemical analysis of adult rat brain found Pitpnm2 prominently expressed in cell bodies within pyramidal layer V and in Purkinje cells of the cerebellum. PITPNM2 expression was also detected in supraoptic nuclei, in the middle of the preoptic region, and in the inner segment and the inner and outer plexiform layers of the retina.
