FAPP2 localized to the Golgi complex in 3 mammalian kidney cell lines. Expression of transfected fluorescence-tagged FAPP2 overlapped specifically with trans-Golgi markers. Deletion of the pleckstrin homology (PH) domain resulted in a cytosolic distribution of FAPP2 in transfected cells.FAPP2 is a glucosylceramide-transfer protein that has a pivotal role in the synthesis of complex glycosphingolipids, key structural and signaling components of the plasma membrane. The requirement for FAPP2 makes the whole glycosphingolipid synthetic pathway sensitive to regulation by PtdIns(4)P and ARF1. Displacement of FAPP1 or FAPP2 from the trans-Golgi network or knockdown of FAPPs with small interfering RNA inhibited cargo transfer to the plasma membrane. Overexpression of the FAPP PH domain also impaired carrier vesicle fission. .
