MyoGEF interacts directly with nonmuscle myosin II and showed that MyoGEF activates RhoA and induces formation of myosin filaments. Both MyoGEF and myosin II colocalize to the cleavage furrow of early anaphase cells. Disruption of MyoGEF expression by RNAi resulted in the formation of multinucleated cells.
The N-terminal domain of ezrin binds to a C-terminal region of PLEKHG6. PLEKHG6 exhibited preferential GEF activity toward RhoG , which may be its primary target. PLEKHG6 occurs in a complex with ezrin, RhoG, and the RhoG effector ELMO. PLEKHG6, together with ezrin, induced ruffles at the cell membrane, and both were necessary for macropinocytosis to occur.The deduced protein showed highest sequence identity with GEF720 (PLEKHG5).
