By database analysis, Pearce et al. (2007) identified a gene encoding a 368-amino acid protein displaying 39% sequence identity with the PROTOR1 protein , which interacts specifically with RICTOR in the mammalian target of rapamycin complex-2 (MTORC2). Antibodies against either PROTOR1 or PROTOR2 resulted in immunoprecipitation of other MTORC2 proteins.The International Radiation Hybrid Mapping Consortium mapped the PROTOR2 gene to chromosome 11.Coexpression of PROTOR1-alpha with MTORC2 subunits (MTOR; RICTOR; and SIN1) demonstrated that PROTOR1 interacts specifically with RICTOR. In Rictor -/- knockout mouse embryonic fibroblast cells, expression of Protor1 and Sin1 was greatly decreased, suggesting that the stability and/or expression of both are dependent on the presence of Rictor.
