By searching EST databases for sequences encoding putative gamma-carboxyglutamic acid (Gla) domain containing proteins, followed by 5-prime and 3-prime RACE of human adult spinal cord cDNA library, Kulman et al. (2001) cloned PRRG3, The deduced 231-amino acid protein lacks an N-terminal signal sequence, but has a propeptide sequence that is cleaved to produce a mature 212-amino acid protein with a Gla domain of up to 13 Gla residues, a single-pass transmembrane domain, and a cytosolic C-terminal domain of 129 amino acids. The cytoplasmic domain contains 2 PPxY motifs that are predicted to interact with WW domain-containing proteins. TMG3 expression in most adult and fetal human tissues. Expression in adult skeletal muscle was decreased relative to fetal muscle, and expression in adult heart was elevated relative to fetal heart.
