The full-length protein has 259 amino acids, and the shorter variant has 211 amino acids with a calculated molecular mass of 23.7 kD. Both isoforms contain 3 phosphate/Mg(2+)-binding motifs, 2 guanosine-binding motifs, an ATP/GTP-binding site, and a myristoylation site. The shorter variant contains a C-terminal microbody targeting signal. The full-length protein lacks this targeting signal but does have 48 additional C-terminal residues. Northern blot analysis of human tissues detected a 1.4-kb RAB34 transcript with high expression in liver, moderate expression in spleen, colon, heart, placenta, lung, skeletal muscle, and kidney, and low expression in all other tissues examined. A minor 2.0-bk transcript was also found in most tissues examined, and a 4.4-kb transcript was detected in spleen and liver.
