RASL11B forms a characteristic RAS GTPase domain with the typical topology of a 6-stranded beta-sheet surrounded by 5 alpha helices. The protein has a putative cleavage site at position 20 but contains no N-terminal signal peptide. Its amino acid sequence predicts a cytoplasmic localization. An RASL11B splice variant encodes a putative 191-amino acid protein that contains the RAS GTPase domain but lacks 57 amino acids in the C terminus that are highly conserved within the RASL11 protein family. Real-time PCR of 37 human tissues and 5 different cell types detected ubiquitous expression of RASL11B but at highly variable levels with differences up to 900-fold. Highest levels of expression were found in placenta, spleen, kidney, ovary, brain, uterus, testis, and cultured primary macrophages.
