The deduced 416-amino acid protein has a calculated molecular mass of 44.5 kD. Northern blot analysis detected a major 3.4-kb transcript and a minor 7-kb transcript highly expressed in skeletal muscle and more weakly expressed in cardiac muscle. Little to no expression was detected in brain, placenta, lung, liver, kidney, and pancreas. RNA dot blot analysis revealed CAPZIP expression in several lymphoid organs, including spleen, thymus, peripheral blood leukocytes, lymph node, and bone marrow. SDS-PAGE of rabbit skeletal muscle detected Capzip at an apparent molecular mass of 70 kD.Hyperosmotic shock and anisomycin, a protein synthesis inhibitor, stimulated phosphorylation of CAPZIP in human cell lines and triggered dissociation of CAPZIP from CAPZ in Jurkat human T cells.
