Using ALG2 as bait in a yeast 2-hybrid screen of a human peripheral blood cDNA library, Montaville et al. (2006) cloned RBM22. The deduced 420-amino acid protein contains a conserved zinc finger of the type Cx8Cx5Cx3H, followed by an RNA-binding domain. The mouse and human RBM22 proteins are identical. Fluorescence-tagged RBM22 localized to the nucleus of transfected mouse fibroblasts.
Knockdown of RBM22 in HeLa cells resulted in mitotic arrest and a cytokinesis defect, but not cell death.Montaville et al. (2006) found that fluorescence-tagged human RBM22 redirected fluorescence-tagged ALG2 from the cytoplasm to the nucleus following coexpression in mouse fibroblasts and zebrafish embryos.
