Ras Association Domain Containing Protein 4 (RASSF4)
RASSF4 is a member of the RAS association (RA) domain family of RAS effectors/tumor suppressors that mediate some of the growth inhibitory properties of the RAS oncogenes. RASSF4 shares 60% and 25% amino acid identity with RASSF2 and RASSF1, respectively, and shares 72% identity with RASSF2 over their respective RA domains.
RASSF4 induced cell death and synergized with KRAS to promote increased RAS-mediated cell death. In human breast tumor cells, RASSF4 induced apoptosis with a 3-fold increase in the number of tumor cells exhibiting activated caspases. A membrane-targeted C-terminal RASSF4 mutant transfected into breast tumor and lung tumor cell lines inhibited cell growth and colony formation.
