ARHGEF10L is a member of the RhoGEF family of guanine nucleotide exchange factors (GEFs) that activate Rho GTPases. The deduced 1,235-amino acid ARHGEF10L protein has a calculated molecular mass of 135 kD and shares 60% and 44% similarity with ARHGEF10 and ARHGEF17, respectively.
ARHGEF10L contains a DH domain, a WD40-like domain, and 2 membrane-spanning sequences. Northern blot analysis of human tissues detected expression of a 5-kb ARHGEF10L transcript in heart, liver, skeletal muscle, kidney, and pancreas. An additional 4-kb variant was detected in heart and skeletal muscle, and a 6-kb variant was detected in liver. ARHGEF10L functions as a RHO-specific guanine nucleotide exchange factor (GEF).
