It contains an N-terminal C3HC4-type RING finger domain, 2 putative myristoylation sites, and several putative phosphorylation sites. Northern blot analysis detected a 2.5-kb transcript in all tissues examined, with highest expression in heart and skeletal muscle. RNF146 mRNA showed similar abundance in all brain regions examined. Northern blot analysis of mouse tissues detected highest Rnf146 expression in heart and lower expression in all other tissues examined, including skeletal muscle. In situ hybridization of mouse brain revealed widespread expression of Rnf146 in the cytosol of neurons, but not in glial cells. Epitope-tagged human RNF146 was detected in the cytosolic fraction of transfected human glioblastoma cells. Database and Northern blot analyses suggested that RNF146 may be alternatively spliced.
