SET domain-containing proteins, such as SMYD2, catalyze lysine methylation. The deduced 433-amino acid protein has a SET domain that is split into 2 segments by a MYND domain, followed by a cysteine-rich domain. Northern blot analysis of mouse tissues detected highest expression in heart, brain, liver, kidney, and ovary. In situ hybridization of day-13.5 mouse embryos showed Smyd2 expression in heart and hypothalamus.the SET domain of mammalian Smyd2 mediated demethylation of lys36 of histone H3, and Smyd2 repressed transcription from an SV40 reporter plasmid. Exogenous expression of Smyd2 in mouse fibroblasts decreased cell growth.SMYD2 helps to maintain low concentrations of promoter-associated p53.
