SH3 Domain GRB2 Like Protein 3 (SH3GL3)
The Src homology 3 domain (SH3) is an evolutionarily conserved, 50- to 60-amino acid module carried by intracellular proteins involved in the transduction of signals for cell polarization, motility, enzymatic activation, and transcriptional regulation. The SH3 drives protein-protein interactions through binding to proline-rich ligands. Coimmunoprecipitations and immunofluorescence studies revealed that SH3GL3 and HDex1p colocalize in transfected COS cells. Additionally, an anti-SH3GL3 antibody was able to coimmunoprecipitate the full-length huntingtin from an HD human brain extract. The characteristics of the interaction between SH3GL3 and huntingtin and the colocalization of the 2 proteins suggested that SH3GL3 could be involved in the selective neuronal cell death in HD.
