Corl1 interacted with Lbx1, a homeodomain transcription factor selectively expressed in dorsal horn interneurons, and cooperated with Lbx1 to repress transcription in a neuroblastoma cell line. Mutation analysis revealed that the N- and C-terminal regions of Corl1 were required for interaction with Lbx1 and that the CH1 domain was required for transcriptional repression. Corl1 corepressor activity did not depend on Grg1, which also functioned as a Lbx1 corepressor.The deduced 936-amino acid protein has an N-terminal cysteine-rich region, followed by a Corl homology (CH1) domain and a C-terminal coiled-coil region. Corl1 was also expressed in a subset of postmitotic neurons posterior to the midbrain-hindbrain border. Epitope-tagged Corl1 localized in nuclear dot-like structures of transfected human embryonic kidney cells.
