Mouse embryonic fibroblasts (MEFs) were transfected with retroviruses, resulting in random retroviral integration in their genomes and then co-inoculated with tumor cells into mice.Genomic analysis of co-inoculated stromal cells recovered from tumors showed clone-specific selection of cells with certain mutations during tumor progression.
Four integration sites were detected after in vivo selection, including Tubulin-tyrosine ligase (TTL), Histone 1, Rhesus blood group-associated B glycoprotein (RHGB) and SSTK-interacting protein (SSTK IP). TTL silencing promoted tumor growth. The retrovirus-mediated insertional mutagenesis may provide a valuable means for identifying genes involved in the interactions between tumor and stromal cells.
