TDE1 overexpression in transfected rat cells resulted in reduced contact inhibition with formation of robust foci. In transfected rat and human cell lines, TDE1 overexpression reduced apoptosis caused by serum starvation or etoposide treatment, as shown by reduced caspase-3 activation and DNA laddering. TDE1 did not alter cell growth rate, immortalization, or motility. The 473-amino acid protein has a predicted molecular mass of 52 kD. Use of an alternative polyadenylation site results in a splice variant lacking 61 amino acids. Like the mouse protein, with which it shares 78% sequence homology, human TDE1 contains several predicted hydrophobic alpha helices characteristic of transmembrane proteins, and it has several potential phosphorylation sites.
