By direct cDNA selection, followed by database analysis and screening of human brain, liver, and testis cDNA libraries, Ruddy et al. (1997) cloned SLC17A3, which they called NPT4. The 401-amino acid protein has 4 potential transmembrane domains. Northern blot analysis revealed that SLC17A3 is expressed in liver and kidney, and RT-PCR analysis showed expression in small intestine, liver, kidney, and testis.
Melis et al. (2004) localized NPT4 expression in COS-1 cells to the endoplasmic reticulum by immunofluorescence.Melis et al. (2004) determined that the SLC17A3 gene contains 10 exons.By genomic sequence analysis, Ruddy et al. (1997) mapped the SLC17A3 gene to chromosome 6p21.3.
