SLC2A12 belongs to a family of transporters that catalyze the uptake of sugars through facilitated diffusion . This family of transporters show conservation of 12 transmembrane helices as well as functionally significant amino acid residues .
By database analysis, followed by RT-PCR of MCF7 cells, they cloned the full-length cDNA. The deduced 617-amino acid protein has a calculated molecular mass of about 67 kD. Northern blot analysis detected 2 major transcripts in MCF7 cells. Examination of several human tissues detected highest expression in heart, skeletal muscle, and prostate, and lower levels in brain, placenta, and kidney. Western blot analysis detected GLUT12 at an apparent molecular mass of about 50 kD. Immunohistochemical analysis detected intense GLUT12 staining in skeletal muscle bundles.
