C4ORF1 encodes a hydrophilic, 568-amino acid protein with a pI of 8.61. The protein contains an LXXLL nuclear receptor interaction motif associated with some transcriptional coactivators, several nuclear localization signals, a nuclear export-like signal, a zinc finger-like motif, an acidic region, and 2 leucine zipper-like domains. Northern blot analysis revealed expression of a 3.4-kb transcript in fetal brain, lung, liver, and kidney. RT-PCR analysis revealed ubiquitous expression in adult tissues as well as cancer cell lines. In immunoblot analyses, affinity purified rabbit antibodies against HUEL recognized a 70-kD protein in lung and hepatoma cell line lysates, close to the calculated mass of 63.4 kD, predominantly in the cytosol. Cytoplasmic staining was also observed in fluorescent antibody studies.
