The deduced 573-amino acid protein contains 83 N-terminal amino acids not found in human SLC41A2. It has 12 transmembrane domains, a putative N-glycosylation site, several possible phosphorylation sites, and 3 myristoylation consensus sequences.
Mouse Slc41a2 mediated voltage-dependent and saturable Mg(2+) uptake when expressed in Xenopus oocytes. Slc41a2 also transported Ba(2+), Ni(2+), Co(2+), Fe(2+), and Mn(2+), but not Ca(2+), Zn(2+), or Cu(2+). Mg(2+) transport was inhibited by high Ca(2+) concentrations. Real-time PCR of mouse renal distal tubule epithelial cells showed that expression of Slc41a2, unlike that of Slc41a1, was not responsive to magnesium.
