Kst1 protein also transported D-chiro-inositol but did not transport L-glucose, L-xylose, or any other sugar examined. Transport was sodium-dependent. Presteady-state currents were phlorizin-inhibitable, and there was a substrate-independent sodium leak current similar to that measured in related cotransporters.SLC5A11 contains 14 transmembrane domains, and it shares significant homology with other members of the sodium/glucose cotransporter family. Skipping of exon 6 results in an alternatively spliced transcript encoding a deduced 640-amino acid protein lacking transmembrane domain 4. Northern blot analysis detected a 5.2-kb transcript with highest expression in heart, skeletal muscle, kidney, liver, and placenta, and weaker expression in brain, spleen, lung, and peripheral blood leukocytes.
