Src Homology 2 Domain Containing Transforming Protein D (SHD)
Using a fragment of human ABL1 as bait in a yeast 2-hybrid screen of an embryonic mouse cDNA library, Oda et al. (1997) cloned Shd. The deduced 343-amino acid protein contains an N-terminal proline-rich region, a C-terminal SH2 domain, and 5 YxxP tyrosine phosphorylation motifs. Shd shares highest similarity with Shb . Northern blot analysis detected Shd expression in mouse brain only.
Oda et al. (1997) found that mouse Shd interacted with kinase-active human ABL1, but not with a kinase-defective ABL1 mutant. Pull-down assays showed that the SH2 domain of Shd bound to several other phosphotyrosine-containing proteins in a human myelogenous leukemia cell line. Shd was phosphorylated by ABL1 following cotransfection in COS-7 cells.
