Svep1-transfected COS-7 cells expressed a protein of more than 400 kD, consistent with N-glycosylation. Northern blot analysis of human tissues detected expression in placenta with little to no expression detected in other tissues. Mouse Svep1 expression was detected at high levels in lung and placenta, and Svep1 mRNA was detectable from day 11 postcoitum to day 17 postcoitum in mouse embryos.SVEP1 shares similarity with the selectin family of adhesion proteins but does not contain a lectin domain. Immunoprecipitation and Western blot analysis detected SVEP1 in mesenchymal primary cultured cell lysates and in murine stromal osteogenic cells. Immunohistochemistry and confocal microscopy localized SVEP1 to the cell membrane and cytoplasm.
