JFC1 interacted with p47-phox (NCF1) indirectly by binding the p47-phox/p67-phox complex; it had no effect on the interaction between p67-phox and p47-phox. The deduced 562-amino acid protein contains a second putative start codon and tandem C2 domains similar to those found in synaptotagmins. Northern blot analysis detected a 1.8-kb transcript in leukocytes. Semiquantitative mRNA dot blot analysis detected abundant expression in bone marrow, lymphoid tissue, pancreas, trachea, stomach, salivary gland, and prostate. Following density gradient fractionation, JFC1 segregated with neutrophil membranes and secretory vesicles, but not with azurophils or other granules. Phorbol ester stimulation did not affect the subcellular distribution, but high salts could partially release JFC1 from its membrane association.
