The deduced 803-amino acid protein contains 4 N-terminal transmembrane domains and 16 C-terminal leucine-rich repeats. LRRC8B shares 59% amino acid identity with LRRC8A and 93% identity with the mouse ortholog, which also contains 803 amino acids. Semiquantitative PCR detected variable LRRC8B expression in all tissues examined except skeletal muscle.The transcript contains an Alu element in its 3-prime end. The deduced protein contains several leucine-rich repeats and shares significant similarity with yeast adenylate cyclase. Northern blot analysis detected low LRRC8B expression in all tissues and cell lines examined.expression of LRRC8B in isolated human peripheral blood mononuclear cells or monocytes was increased following exposure to phytohemagglutinin, phorbol ester, or lipopolysaccharide.
