Markk phosphorylated Mark on a threonine within its activation loop. In brain, Markk also phosphorylated a fraction of Mark on a nearby serine, and this phosphorylation inhibited Mark activity. In cells, Markk activity enhanced microtubule dynamics through activation of Mark and led to phosphorylation and detachment of microtubule-associated proteins from microtubules. The cDNA contains repetitive elements in its 5-prime and 3-prime regions, and the deduced 1,005-amino acid protein shares 99% identity with rat Tao1. Intermediate expression was detected in all other brain regions and adult and fetal tissues examined, with slightly higher levels in caudate nucleus, substantia nigra, whole adult brain, adult liver, and fetal liver.
