The C-terminal domains of TBPL2 and TBP share 93% identity, but their N-terminal domains are nonconserved. The N-terminal domains of vertebrate TBPL2 sequences are relatively divergent, except for residues 98 to 104, which are identical from fish to human. HeLa cell TBPL2 showed an apparent molecular mass of about 55 kD, and gel filtration chromatography indicated that TBPL2 is a component of a multisubunit complex of about 200 kD. Immunofluorescence analysis of asynchronized HeLa cells found TBPL2 localized in the nucleus. In synchronized cells, TBPL2 moved to the cytoplasm during cytokinesis in late M phase, and it was virtually undetectable by S phase. After mitosis, TBP and other transcription factors were reimported into the nucleus before TBPL2.
