T3jam contains 513 amino acids. By database analysis, they identified human T3JAM, which encodes a deduced 531-amino acid protein that is 66% identical to the mouse protein. T3JAM contains a coiled-coil domain homologous to the myosin tail heptad repeat and the ezrin -radixin-moesin (ERM) family domain. Western blot analysis of mouse tissues detected T3jam expression in bone marrow, spleen, and thymus, but not in heart, kidney, or liver.Dadgostar et al. (2003) determined that the coiled-coil domain of mouse T3jam interacted with the isoleucine zipper domain of Traf3. T3jam did not associate with other Traf family members. Coexpression of T3jam and Traf3 recruited Traf3 to the detergent-insoluble fraction, and T3jam and Traf3 synergistically activated JNK, but not nuclear factor kappa-B.
