TRRP4 Associated Protein (TRPC4AP)

TRRP4AP; TRUSS; C20orf188; Transient Receptor Potential Cation Channel Subfamily C Member 4 Associated Protein; TNF-receptor ubiquitous scaffolding/signaling protein

TRRP4 Associated Protein (TRPC4AP)
Trpc4ap, the deduced 797-amino acid protein has a calculated molecular mass of about 90.7 kD. It contains 2 putative Traf2-binding sites, several putative phosphorylation sites, and 10 C-terminal dilucine motifs. Northern blot analysis detected a 3.2-kb transcript widely expressed in mouse tissues, with highest levels in heart, brain, liver, and testis. Western blot analysis detected a protein of about 82 kD expressed in several mouse cell lines and in a human promonocytic cell line.Transient overexpression of Truss activated nuclear factor kappa-B (NFKB) and increased Nfkb activation in response to ligation of Tnfr1. In contrast, a C-terminal deletion mutant of Truss inhibited Nfkb activation. Coprecipitation and coimmunoprecipitation assays revealed that Truss interacted with Tradd , Traf2, and components of the IKK complex.
