Transmembrane Emp24 Protein Transport Domain Containing Protein 10 (TMED10)
TMED10 is a member of the EMP24/GP25L/p24 family and encodes a protein with a GOLD domain. This protein is localized to the plasma membrane and golgi cisternae and is involved in vesicular protein trafficking. The protein is also a member of a heteromeric secretase complex and regulates the complex's gamma-secretase activity without affecting its epsilon-secretase activity. Mutations in this gene have been associated with early-onset familial Alzheimer's disease. The deduced 219-amino acid type I intracellular transmembrane protein contains a signal sequence and is predicted to be located in the lumen of the endoplasmic reticulum. Northern blot analysis detected a 1.4-kb TMP21 transcript. Immunoblot analysis showed that the rat Tmp21 protein is expressed predominantly in the microsomal fraction of pancreatic acinar cells.
