Both the mouse and human proteins contain 477 amino acids and have an N-terminal tripartite motif consisting of RING, B-box, and coiled-coil domains, followed by a C-terminal SPRY domain.
Fluorescence-tagged Mg53 and endogenous Mg53 localized to the sarcolemmal membrane and intracellular vesicles of mouse skeletal muscle.Scrape-wounding of C2C12 cells resulted in dense accumulation of fluorescence-tagged Mg53 at the site of membrane damage, and this accumulation required Mg53 oligomerization and was associated with nucleation of vesicles into a membrane patch. Oligomerization of Mg53 required oxidation of cys242, and interaction of Mg53 with phosphatidylserine mediated recruitment of vesicles to sites of injury. .
