GALNT12 is a member of a family of UDP-GalNAc:polypeptide N-acetylgalactosaminyltransferases (EC 2.4.1.41), which catalyze the transfer of N-acetylgalactosamine (GalNAc) from UDP-GalNAc to a hydroxyl amino acid on a polypeptide acceptor in the initial step of mucin-type O-linked protein glycosylation .
The deduced 581-amino acid protein contains an N-terminal cytoplasmic domain, transmembrane domain, and putative stem region. The predicted GALNT12 catalytic domain contains a GT1 motif, Gal/GalNAc transferase motif, and ricin-like lectin motif common to other GalNAcT family members. GALNT12 shares 32 to 56% amino acid sequence homology with other GalNAcT family members with highest homology to GALNT4.
