The deduced 379-amino acid protein has a calculated molecular mass of 42 kD. It has strong similarity to the UB-binding domain of E2 UB-conjugating enzymes in its N-terminal half, but, like TSG101, a cysteine required for UB conjugase activity (cys101) is replace with tyrosine, likely inactivating the UB-binding domain.
At its C terminus, UEV3 shares significant similarity to lactate dehydrogenases. Northern blot analysis detected 2.5- and 4.5-kb UEV3 transcripts in all colon carcinoma cell lines examined. RT-PCR showed comparable expression of UEV3 in colon carcinomas and corresponding normal colon mucosa, and a UEV3 variant lacking exon 3 was also present in all samples.UEVLD gene contains 11 exons and spans 56 kb.
