Proteins that contain a FYVE domain, such as WDFY2, bind with high specificity and avidity to phosphatidylinositol 3-phosphate (PtdIns3P).
The deduced protein contains 7 WD40 repeats with a FYVE domain inserted between repeats 6 and 7. Immunohistochemical analysis of COS-7 cells detected endogenous Wdfy2 staining of vesicular structures of various sizes distributed over the entire cytoplasmic volume, but Wdfy2 did not colocalize with classical endosomal markers.treatment of WDFY2-transfected COS-7 cells with wortmannin, a PI3K inhibitor, resulted in relocalization of WDFY2-containing endosomes. Knockdown of WDFY2 in HeLa cells with siRNA resulted in inhibited transferrin receptor (TFRC) internalization.
