The WD40 repeat domain is a common structural module in eukaryotes, and proteins containing WD40 domains have a diverse range of functions, including signal transduction, cell cycle regulation, RNA splicing, and transcription. Both human and mouse MONAD encode a 357-amino acid protein, and immunoblot analysis of rat cortex with human MONAD antibody detected the protein at 40 kD. MONAD contains only 2 WD40 repeats, in contrast to other known WD40 repeat proteins that contain 4 or more repeats. Quantitative PCR showed highest expression in testis and moderate expression in heart, kidney, placenta, pancreas, and skeletal muscle.overexpression of MONAD in HEK293 cells potentiated apoptosis and caspase-3 activation induced by TNF-alpha and cyclohexamide.
