The deduced 622-amino acid protein contains 5 ankyrin repeats in its N-terminal half and 6 transmembrane domains in its C-terminal half. HIP14 and HIP14L share 48% amino acid identity.RT-PCR analysis showed that both Hip14 and Hip14l were upregulated by low Mg(2+) concentrations. Low Mg(2+) increased expression of fluorescence-tagged HIP14L in canine kidney cells, and it increased recruitment of HIP14L to subplasma membrane vesicles. Mouse or human cRNAs for HIP14 and HIP14L mediated Mg(2+) transport following expression in Xenopus oocytes, and both proteins localized predominantly on the surface. Mg(2+) uptake was electrogenic, voltage dependent, and saturable. Treatment of HIP14 and HIP14L with a palmitoylation inhibitor, or deletion of the DHHC motif in HIP14, reduced Mg(2+) transport.
