Zinc Finger Protein 274 (ZNF274)
ZNF274 encodes a zinc finger protein containing five C2H2-type zinc finger domains, one or two Kruppel-associated box A (KRAB A) domains, and a leucine-rich domain. The encoded protein has been suggested to be a transcriptional repressor. It localizes predominantly to the nucleolus. Alternatively spliced transcript variants encoding different isoforms exist. These variants utilize alternative polyadenylation signals.The deduced 621-amino amino acid ZNF274A protein has 5 C2H2-type zinc finger motifs with 2 KRAB A domains separated by an LRR domain. In contrast, the deduced 584-amino acid ZNF274B protein has only 1 KRAB A domain. Northern blot analysis revealed wide expression of 2.5- to 3.0-kb transcripts at varying levels. Reporter gene analysis indicated that the KRAB domain of ZNF274 has transcription-repressing activity.
