[PRODUCT INFORMATION] 

Streptavidin is a protein secreted by streptomyces avidinii. It exists in the form of homologous tetramer, which can highly specific binding 4 moles of biotin molecules per mole, and its non-specific binding rate is much lower than that of avidin. So, it can be widely used in various signal amplification systems for biochemical and immune detection. Streptavidin-agarose beads is a kind of microsphere resin immobilized recombinant streptavidin protein, it can be used for the capture, purification and immunoprecipitation of biotinylated proteins, antibodies, nucleic acids and other molecules.

[APPLICATION] 

Streptavidin-agarose beads can be used for the capture, purification and immunoprecipitation of biotinylated molecules. 


[METHOD] 

The following is a general method for purifying biotinylated proteins. It is suggested to determine the optimal buffer system and experiment conditions depend on different protein types. 

Buffer preparation: 

Binding buffer:0.01M PBS, pH 7.2~7.4. 

Elution buffer:100mM glycine, pH 2.2~2.8. 

  1. Filling streptavidin-agarose microsphere resin (suitable for filling various medium pressure chromatographic columns).

  2. Connect the prepared chromatographic column to the chromatographic system. Balance the resin with a binding buffer of 5 times the column volume.

  3. The biotinylated protein samples were added to the chromatographic column. Ensure the samples were in full contact with the resin, and incubated at room temperature for 1~2h.

  4. Clean the chromatographic column with binding buffer of at least 10 times the column volumes, to remove non-specific adsorbed proteins.

  5. Elute the bound biotinylated samples with elution buffer of 5-10 times the column volumes.

  6. Use suitable buffer to dialyze the eluted samples according to downstream application, and preserve the purified samples. 

  7. Clean and store streptavidin agarose microsphere resin.

[STORAGE AND EXPIRATION DATE] 

Unopened product stored at 4℃ for 12 months; Store at 4℃ for 1-2 weeks after opening and use as soon as possible. It is recommended to add 0.03% proclin300 during storage for repeated use.
