The MECT1 mRNA contains a repetitive element in the 3-prime UTR. Following in vitro translation, the deduced 634-amino acid protein had an apparent molecular mass of 75 kD by SDS-PAGE. expression in brain and kidney, intermediate expression in heart, lung, liver, skeletal muscle, pancreas, testis, and ovary, and little to no expression in spleen. Torc1 was expressed in adult mouse brain and cultured neurons, and that it translocated to the nucleus upon concomitant activation of calcium and cAMP signaling pathways. Nuclear translocation of Torc1 resulted in synergistic activation of Creb-mediated transcription, and Torc1 played a role in activation of brain-derived neurotrophic factor (BDNF) by calcium and cAMP. Torc1 was involved in late-phase long-term potentiation maintenance at the Schaffer collateral-CA1 synapses in the hippocampus.
